Hydrocodone, chemically known as dihydrocodeinone, is a semi-synthetic opioid primarily utilized for pain management and as a cough suppressant. It is administered orally and is often found in combination formulations, such as acetaminophen/hydrocodone or ibuprofen/hydrocodone, designed for individuals experiencing significant pain that necessitates opioid treatment. Additionally, it can be combined with homatropine methylbromide to alleviate cough symptoms. A long-acting version, marketed under the brand names Hysingla ER, Zohydro ER, Vantrela ER and Vicodin ES are also available for the management of chronic severe pain.

First patented in 1923, hydrocodone's extended-release formulation received approval for medical use in the United States in 2013. The drug is predominantly prescribed in the U.S., which accounted for 99% of global consumption as of 2010. By 2018, hydrocodone ranked as the 402nd most frequently prescribed medication in the country, with over 400,000 prescriptions issued. This semi-synthetic opioid is derived from codeine, and less commonly, from thebaine. Although methods for producing hydrocodone using genetically modified yeasts have been developed, they are not yet employed in commercial production.
